Amivantamab plus Lazertinib vs Osimertinib plus Chemotherapy for the First-Line Treatment of Advanced Non-Small Cell Lung Cancer

The AIHTA assessment found no confirmed clinical benefit of amivantamab plus lazertinib compared with osimertinib plus chemotherapy for advanced NSCLC with common EGFR mutations. At the same time, the therapy is associated with higher costs and additional organisational requirements.

Balance scale with medicines and laboratory equipment on both sides.

The AIHTA assessed the clinical, health economic and organisational impacts of amivantamab plus lazertinib compared with osimertinib plus platinum-based chemotherapy as a first-line treatment for advanced non-small cell lung cancer (NSCLC) with common EGFR mutations.

The currently available evidence does not demonstrate a proven clinical benefit of amivantamab plus lazertinib over osimertinib plus chemotherapy. Although overall rates of severe adverse events are similar between the two regimens, their toxicity profiles differ qualitatively. Amivantamab plus lazertinib is characterised by skin toxicity and venous thromboembolism, while osimertinib plus chemotherapy causes a temporary reduction in blood cell production in the bone marrow. This can increase the risk of infections, fatigue, and bleeding.

At the same time, amivantamab plus lazertinib is associated with higher costs and additional organisational requirements. Although the subcutaneous formulation of amivantamab reduces the burden of administration, the requirements for monitoring and supportive care largely remain unchanged. However, the evidence base continues to be affected by uncertainty due to the lack of direct comparative studies.
